The British actor - who has previously played ex-British prime minister Tony Blair - will take the role of Aro, leader of an Italian vampire clan called the Volturi in the film.
The film's director, Chris Weitz, said he "aggressively" pursued Michael to appear in the film, which also stars Robert Pattinson and Dakota Fanning.
Chris said: "Michael's role is so important because he's the head of all vampires. Aro is, on the surface, a very gracious and friendly vampire, but beneath that he is a tremendous threat."
Michael rose to fame portraying Tony Blair in 'The Queen' and recently appeared as British journalist David Frost in 'Frost/Nixon' and football manager Brian Clough in 'the Damned United'. He has previously played werewolf Lucian in the 'Underworld' trilogy, the latest of which 'Underworld: Rise of the Lycans' was released in January this year
'The Twilight Saga: New Moon' is set for release in November.
